The former number 1 player in the world began with four straight birdies, and finished with birdies on both par 5s for a 6-under 65 and a one-shot lead in the Marathon Classic.
Ko has gone 44 tournaments without winning, and has slipped to number 55 in the world.
Jodi Ewart Shadoff shot 63 and was one shot behind.
Danielle Kang, seeking her second straight victory in Ohio after winning last week at Inverness, shot 67 and was two shots behind.
Princeton's Emma Talley shot a 5-under 66 on Friday and is tied for 19th. She's 4 under for the tournament, nine shots off the lead.
